RESUMO

Epigenetic modifications play a vital role in the preservation of genome integrity and in the regulation of gene expression. DNA methylation, one of the key mechanisms of epigenetic control, impacts growth, development, stress response and adaptability of all organisms, including plants. The detection of DNA methylation marks is crucial for understanding the mechanisms underlying these processes and for developing strategies to improve productivity and stress resistance of crop plants. There are different methods for detecting plant DNA methylation, such as bisulfite sequencing, methylation-sensitive amplified polymorphism, genome-wide DNA methylation analysis, methylated DNA immunoprecipitation sequencing, reduced representation bisulfite sequencing, MS and immuno-based techniques. These profiling approaches vary in many aspects, including DNA input, resolution, genomic region coverage, and bioinformatics analysis. Selecting an appropriate methylation screening approach requires an understanding of all these techniques. This review provides an overview of DNA methylation profiling methods in crop plants, along with comparisons of the efficacy of these techniques between model and crop plants. The strengths and limitations of each methodological approach are outlined, and the importance of considering both technical and biological factors are highlighted. Additionally, methods for modulating DNA methylation in model and crop species are presented. Overall, this review will assist scientists in making informed decisions when selecting an appropriate DNA methylation profiling method.

The family of Nuclear Distribution C (NudC) proteins plays a pivotal and evolutionarily conserved role in all eukaryotes. In animal systems, these proteins influence vital cellular processes like cell division, protein folding, nuclear migration and positioning, intracellular transport, and stress response. This review synthesizes past and current research on NudC family members, focusing on their growing importance in plants and intricate contributions to plant growth, development, and stress tolerance. Leveraging information from available genomic databases, we conducted a thorough characterization of NudC family members, utilizing phylogenetic analysis and assessing gene structure, motif organization, and conserved protein domains. Our spotlight on two Arabidopsis NudC genes, BOB1 and NMig1, underscores their indispensable roles in embryogenesis and postembryonic development, stress responses, and tolerance mechanisms. Emphasizing the chaperone activity of plant NudC family members, crucial for mitigating stress effects and enhancing plant resilience, we highlight their potential as valuable targets for enhancing crop performance. Moreover, the structural and functional conservation of NudC proteins across species suggests their potential applications in medical research, particularly in functions related to cell division, microtubule regulation, and associated pathways. Finally, we outline future research avenues centering on the exploration of under investigated functions of NudC proteins in plants.

Treatment of complex osteoporotic distal femur fractures with the Less Invasive Stabilization System (LISS) has been associated with high complication rates. The aim of this study was to investigate the biomechanical competence of two different techniques of augmented versus conventional LISS plating. Unstable distal femoral fracture AO/OTA 33-A3 was created via osteotomies in artificial femora simulating osteoporotic bone. Three study groups, consisting of 10 specimens each, were created for fixation with either LISS plate, LISS plate with additional polylactide intramedullary graft, or LISS plate plus medial locking plate (double plating). All specimens were non-destructively tested under axial (20-150 N) and torsional (0-4 Nm) quasi-static loading. Each bone-implant construct was tested with two different working length (WL) configurations (long and short) of the LISS plate. Relative movements between the most medial superior and inferior osteotomy aspects were investigated via three-dimensional motion tracking analysis. Double plating revealed significantly smaller longitudinal and shear displacement than the other two techniques (p ≤ 0.001). In addition, LISS plus graft fixation was with significantly less longitudinal displacement in comparison to conventional LISS plating (p < 0.001). Long WL resulted in significantly higher longitudinal and shear displacement compared to short WL for LISS and LISS plus graft (p ≤ 0.032), but not for double plating (p > 0.999). In conclusion, intramedullary grafting resulted in significantly increased fracture stability under axial loading in comparison to conventional LISS plating. Although it was not efficient enough to provide comparable stability to double plating, intramedullary grafting may be considered as a useful biological alternative to the latter in a surgeon's armamentarium. © 2018 Orthopaedic Research Society. In this paper we present the results of a four-year study of water quality in runoff from an extensive, sedum covered, vegetated roof on an urban commercial building. Monitoring commenced seven months after the roof was constructed, with the first growing season. Stormwater drainage quality function of the vegetated roof was compared to a conventional (impermeable, high-albedo) membrane roof in addition to paired measurements of wet and bulk depositions at the study site. We present concentrations and fluxes of nutrients and major solutes. We discuss seasonal and year-to-year variation in water quality of drainage from the vegetated roof and how it compares with atmospheric deposition and drainage from the impermeable roof. Drainage waters from the vegetated roof exhibited a high concentration of nutrients compared to atmospheric deposition, particularly during the warm temperature growing season. However, nutrient losses were generally low because of the strong retention of water by the vegetated roof. There was marked variation in the retention of nutrients by season due to variations in concentrations in drainage from the vegetated roof. The vegetated roof was a sink of nitrogen, total phosphorus and chloride, and a source of phosphate and dissolved inorganic and organic carbon. Chloride exhibited elevated inputs and leaching during the winter. The drainage from the vegetated and impermeable roofs met the United States Environmental Protection Agency freshwater standards for all parameters, except for total phosphorus.

Syracuse, New York is working under a court-ordered agreement to limit combined sewer overflows (CSO) to local surface waters. Green infrastructure technologies, including green roofs, are being implemented as part of a CSO abatement strategy and to develop co-benefits of diminished stormwater runoff, including decreased loading of contaminants to the wastewater system and surface waters. The objective of this study was to examine the quantity and quality of discharge associated with precipitation events over an annual cycle from a green roof in Syracuse, NY and to compare measurements from this monitoring program with results from a roof irrigation experiment. Wet deposition, roof drainage, and water quality were measured for 87 storm events during an approximately 12 month period over 2011-2012. Water and nutrient (total phosphorus, total nitrogen, and dissolved organic carbon) mass balances were conducted on an event basis to evaluate retention annually and during the growing and non-growing seasons. These results are compared with a hydrological manipulation experiment, which comprised of artificially watering of the roof. Loadings of nutrients were calculated for experimental and actual storms using the concentration of nutrients and the flow data of water discharging the roof. The green roof was effective in retaining precipitation quantity from storm events (mean percent retention 96.8%, SD = 2.7%, n = 87), although the relative fraction of water retained decreased with increases in the size of the event. There was no difference in water retention of the green roof for the growing and non-growing seasons. Drainage waters exhibited high concentration of nutrients during the warm temperature growing season, particularly total nitrogen and dissolved organic carbon. Overall, nutrient losses were low because of the strong retention of water. However, there was marked variation in the retention of nutrients by season due to variations in concentrations in roof runoff.

We quantified internal processes that supply methylmercury from hypolimnetic reducing zones to the upper waters of a Hg-contaminated lake, Onondaga Lake, NY, USA. Diffusive transport continuously supplied methylmercury to the epilimnion under summer stratification, while fall mixing resulted in a pulsed release of methylmercury to the upper mixed waters. These processes were the main internal sources of methylmercury to the epilimnion, and together almost equaled the total external supply. The wind-driven entrainment represented an additional stochastic internal supply of methylmercury of approximately 9% in 2006. Considering more than 15 years of data, we estimate 1.8 wind-driven events occur per year. The mass of methylmercury inputs to the epilimnion exceeded the measured increase, suggesting that loss processes are important in regulating methylmercury accumulation. The relative contribution of internal sources of methylmercury to the epilimnion has decreased in recent years, shifting the importance to the external inputs.

Platinum (II) complexes with cyclobutanespiro-5'-hydantoin and cycloheptanespiro-5'-hydantoin were synthesized and evaluated by means of general physicochemical methods. The data from the elemental analysis, IR and NMR spectra suggested the formation of cis-[Pt(C6H8N2O2)2(NH3)2](NO3)2 x 4H2O (PtCBH), when cyclobutanespiro-5'-hydantoin was used as a ligand and cis-[Pt(C9H14N2O2)(NH3)2](NO3)2 x 4H2O (PtCHTH), when cycloheptanespiro-5'-hydantoin was used, respectively. The novel complexes exerted cytotoxic effects at micromolar concentrations against a panel of human tumor cell lines. They were found to trigger apoptosis in HL-60 and BV-173 cells as evidenced by DNA-laddering detection. The evaluation of the effects of PtCBH, PtCHTH and the antineoplastic drugs cisplatin and oxaliplatin against cultured murine kidney epithelial cells revealed that the hydantoin complexes were far less nephrotoxic in vitro.
